Manipulating gene expression in hepatocytes at a spatial level involves targeting specific zones within the liver lobule, such as periportal and pericentral regions, to study their distinct roles in hepatic function and disease. This approach enables precise dissection of metabolic and regulatory processes that are zonally distributed, such as gluconeogenesis, lipogenesis, and detoxification. The student's role in this project will focus on the comprehensive development and application of adeno-associated viral (AAV) vectors for targeted gene expression in the specific zone of mouse liver. The responsibility will include vector design, generation, purification, and validation in mouse liver. By engaging in these experiments, the student will develop hands-on expertise in molecular biology, viral vector technology, and liver-targeted gene delivery, as well as gain insights into hepatocyte zonation and liver function.
