My lab employs high-dimensional analyses (CyTOF, single cell RNA-seq) to define the phenotypes and functions of T cell populations that are expanded in patients with autoimmune diseases including lupus and rheumatoid arthritis. Our recent work published in Nature described a pathologically expanded CD4+ T cell population called T ‘peripheral helper' (Tph) cells that augments B cell responses within inflamed peripheral tissues.

We have generated large datasets by CyTOF (mass cytometry) that analyze lymphocytes in blood and tissue samples from patients with rheumatoid arthritis, lupus, and other inflammatory diseases. We have opportunities available for students to analyze these datasets using both standard flow cytometric techniques and multi-dimensional analyses with these aims:

  1. To identify lymphocyte populations that are significantly expanded in patients with lupus or rheumatoid arthritis.
  2. To define the most specific cytometric characteristics for these populations.
  3. To analyze changes in the frequency of these cell populations over time in longitudinal samples.

A familiarity with flow cytometry would be helpful but not essential. A very positive outcome would be to identify new populations of lymphocytes altered in autoimmunity that can be sorted for detailed functional and RNA-seq transcriptomic studies. This work is part of a larger effort in the lab to identify new therapeutic targets in autoimmunity and to develop cellular biomarkers to predict response to treatment.
