Overcoming mutational activation of K-Ras is the most pressing issue in medical oncology today. Emerging epidemiological data suggest that the different activating mutations in K-Ras exhibit different clinical behaviors. An unanswered question is whether this has to do with intrinsic biological differences between mutant forms of K-Ras, or some other aspect of cancers with K-Ras mutations. Through large scale bioinformatic analysis, we have found that each K-Ras allele is associated with a distinct group of secondary mutations. This project addresses whether the secondary mutations interact with K-Ras in an allele-specific manner. At a technical level, the project involves engineering cells or primary organoids to express different K-Ras alleles and then performing genetics screens. The major question will be whether each K-Ras allele responds different to perturbation of secondary genes and whether the results of the screen overlap with the genetics of primary cancers.
