The principal aims of this project are to:

  • Investigate the molecular signals that affect the proliferation and neurogenetic behavior in culture
  • Analyze mice carrying mutations of genes of interest

The student will be involved in using primary retinal neuron and glial cell cultures, tissues sections, and mouse behavior assays to evaluate the effects of genetic changes on retinal neuron differentiation, survival, function, and/or disease development.

Glaucoma is the most frequent neurodegenerative disease and a leading cause of blindness, affecting retinal ganglion cells (RGC) and the optic nerve. Current therapies targeted solely at lowering intraocular pressure (IOP) that slows down disease progression but does not halt the vision loss. Our recent reports indicate that neural damage in glaucoma is associated with retinal infiltration of autoreactive T cells which are pre-sensitized by commensal microflora. Mice raised in the absence of commensal microflora do not manifest RGC and optic nerve damage under high IOP. Current project proposes to develop novel glaucoma therapy by evaluating immune-based strategies or candidate drugs. Students will be involved in applying in vitro and/or in vivo models of glaucoma to examine the efficacy and safety of small molecule or biologic drug candidates. Alternatively, student may also be involved in evaluating T cell responses in human patients through our collaboration with glaucoma specialist.

The role of the student is to identify the niche factors that regulate the regenerative potential of endogeneous retinal progenitors in adult mice, using retinal stem cell cultures as well as mice carrying genetic mutations of neurogenic signals.
