Mutations in more than 100 genes cause inherited deafness. Some (nonsyndromic) cause only deafness; others (syndromic) cause both deafness and additional pathology such as blindness. We are developing methods to use gene therapy to treat certain forms of both syndromic and nonsyndromic deafness. There are a variety of challenges in these projects. In one, we must identify both AAV viral capsids and specific gene regulatory elements that will drive expression in the appropriate cell types in the inner ear. In another, the challenge is to fit a long coding sequence into an AAV vector or vectors.The work employs three animal models—mice, zebrafish and nonhuman primates—and involves methods ranging from surgical delivery of vector, to molecular biology for virus production, to physiology and electron microscopy for assessing functional rescue. Potential projects within this program could range from 4 to 12 months.

Two or three of every thousand newborn children suffers from impaired hearing, and about 70% of these cases have genetic causes. Over 100 different genes can cause hearing loss if mutated. We are developing a variety of methods to treat hereditary deafness using gene therapy. We have focused on two forms: one that causes only deafness, and another that causes congenital deafness and progressive blindness. In some cases, we deliver a normal copy of the mutant gene; in others we deliver DNA editing enzymes to correct the mutation. Our goal is to develop efficient and nontoxic AAV vectors that are therapeutic in mouse models, and test them for delivery and toxicity in non-human primates as a route to clinical trials. There is much work to be done, and many potential projects: We need to develop short promoters for specific cell types, test novel viral capsids for efficacy and specificity, design efficient gene editors, and assess rescue of hearing and vision with physiology and microscopy.

Depending on the project, required skills include molecular biology, small animal surgery, histology and microscopy. Day-to-day mentorship will be by senior research fellows with weekly remote meetings with Dr. Corey. Our laboratory is in the Department of Neurobiology at HMS, where the research would be carried out in person.
